Rok 1936. Archeolog Indiana Jones (Harrison Ford) otrzymuje tajne zadanie od służb specjalnych. Według danych wywiadu niemieccy naukowcy odkryli na pustyni starożytne miasto, gdzie w podziemiach świątyni ukryta jest Arka Przymierza. By nie dopuścić do przejęcia cennego zabytku przez nazistów, Jones wyrusza do Egiptu. Podróżując przez Nepal, doktor spotyka swoją dawną sympatię Marion (Karen Allen), której ojciec - także archeolog - odnalazł laskę Mojżesza. Tam zapisane są wskazówki niezbędne do odnalezienia Arki. Niebawem Indy wraz z dziewczyną przybywa do Kairu. Na miejscu badacz przekonuje się, że Niemcy są już bliscy sukcesu i zrobią wszystko, by wyeliminować rywali.
